I work at the interface between academia, industry and clinical practice to discover targets for immune enhancement during ageing. I have pioneered the study of human immunology, focussing on mechanisms that regulate end-stage differentiation of human and mouse T cells. My group developed new technology to study antigen specific immunity in the skin, identifying that excessive inflammation during ageing inhibits immunity. Blocking inflammation with an anti-inflammatory drug (p38 MAPkinase inhibitor) or by vitamin D3 supplementation enhanced immune responsiveness. We also identified a novel inhibitory protein complex in aged T cells that in humans and mice that is regulated by sestrins. Disruption of this complex enhances immune responses in vivo that is now an anti-immune ageing drug target. I have therefore identified novel molecular mechanisms that inhibit immune responses during ageing, the blocking of which enhances tissue specific immunity.
